    My project is focused on the study of the cell wall organization to the yeast Malassezia restricta that is the most prominent yeast found on the human scalp.

    Skills acquired:

    Microbiology:
    •Culture of fungi/ bacteria: E. coli, S. platensis, S. epidermidis, P. acnes, P. pastoris, A. fumigatus and culture optimization and conservation for M. restricta and other Malassezia species
    •Coculture of bacteria/fungi: Development of synthetic medium for M. restricta/ P. acnes/ S. epidermidis.
    Electronic Microscopy:
    •Scanning electron microscopy: Fixation and embedding optimization for M. restricta and P. acnes
    •Transmission electron microscopy: Samples preparation for M. restricta and P. acnes
    Biochemistry:
    •Fungal cell wall/cell membrane analysis: Polysaccharides/lipids analysis by chemical and enzymatic approachs
    •Enzymology: kinetics and inhibition assays, lipid peroxide detection.
    •Separation techniques for proteins, lipids and sugars: chromatography techniques such as affinity, size exclusion, ion-exchange, thin-layer (TLC), gas or liquid chromatography coupled to mass spectrometry ; electrophoresis
    •Analytical techniques: UV-visible spectroscopy, circular dichroism, micro-calorimetry. Flow cytometry setup for clinical samples (BactiFlow), cell hydrophobicity (Goniometer, MATHS test)
    •Protein purification in Pichia pastoris and E.coli
